1Two Unrecoreded Species Belonging to Penicillium Section Exilicaulis in South Korea

Penicillium in section Exilicaulis is characterized by non-vesiculate monoverticillate and biverticillate stipes. Species in sect. Exilicaulis are commonly found in soil and plants in terrestrial environments; however, only a few species have been reported in Korea. To investigate the diversity of Penicillium sect. Exilicaulis, Penicillium species were isolated from terrestrial and marine environments. Based on sequence analyses of β-tubulin, calmodulin, and the second largest subunit of RNA polymerase II loci, 19 strains of Penicillium in sect. Exilicaulis were identified as P. citreonigrum, P. citreosulfuratum, P. corylophilum, P. menonorum, P. rubefaciens, P. velutinum, Penicillium sp. 1, and Penicillium sp. 2. Two of them, P. citreonigrum and P. citreosulfuratum, were confirmed to be new to Korea. Molecular phylogenies and detailed descriptions of the two unrecorded species are provided.

2Morphological and phylogeny of Plenodomus sinensis and P. collinsoniae , two unreported species isolated from soil in Korea

Two unreported fungal isolates, KNU-GW1901 and KNU-AP100C, were collected from soil sample in Gyeongsangbuk-do, Korea. Their cultural and morphological characteristics were examined after 4 weeks of incubation at 25℃ on potato dextrose agar (PDA), malt extract agar (MEA), and oatmeal agar (OA). The conidial shape of KNU-GW1901 was aseptate, hyaline, globose to ellipsoidal, oblong, and reniform to pyriform and 2.61-4.97×1.93-3.61 μm in size, whereas no conidial structures were observed in KNU-AP100C. The internal transcribed spacer (ITS) regions, large subunit (LSU), and small subunit (SSU) sequences were used to determine the taxonomic positions of the strains using the maximum likelihood phylogenetic tree. The isolate KNU-GW1901 was closely clustered with Plenodomus sinensis MFLUCC 17- 0767, and KNU-AP100C was closely matched with P. collinsoniae CBS 120227. Based on the findings of morphological, cultural, and phylogenetic analysis, the isolates KNU-GW1901 and KNU-AP100C were identical to the previously described P. sinensis and P. collinsoniae isolates, respectively, which are first reported in Korea.

3A New Record on Umbelopsis vinacea and Mucor hiemalis f. corticola Isolated from Korea

In the screening of fungal diversity, two strains were collected from the soil of Yeongcheon and dissected guts from the bodies of Chinese rice grasshopper (Oxya chinensis), Chinese grasshopper (Acrida cinerea), and Far eastern devil grasshopper (Oedaleus infernalis) from Daejeon in Korea. They were identified as Umbelopsis vinacea (KNU-YC-1801B) and Mucor hiemalis f. corticola (KNU-20F7, KNU-20F8, KNU-20F9). Multigene phylogenetic analyses of the internal transcribed spacer (ITS) regions, and large subunit (LSU) sequence data confirmed two unreported taxa along with their morphology. The results of molecular phylogeny firmly supported the detailed description and illustration for each taxon. As far as we know, both Umbelopsis vinacea and Mucor hiemalis f. corticola are the first reported taxa in Korea.

Spores of arbuscular mycorrhizal fungi (phylum Glomeromycota) were isolated from rhizosphere soil collected in Korea. We identified the morphological characteristics of the spores and performed a phylogenetic analysis using the rDNA 18S, 5.8S, and 28S regions. To the best of our knowledge, we confirm the presence of three species of glomeromycotan fungi previously not reported in Korea, namely Acaulospora delicata, Dentiscutata colliculosa, and Racocetra alborosea. We described the morphological characteristics and results of phylogenetic analysis of these species.

Endophytic fungi were isolated from the leaves of plants inhabiting the Hansando Island in Korea. We identified the fungal strains based on their morphological characteristics and on the phylogenetic analysis using DNA sequences of the internal transcribed spacer, small subunit rDNA, large subunit rDNA, and translation elongation factor 1- α regions. In this study, we isolated five endophytic fungi that have not been recorded in Korea, Arthrinium camelliae-sinensis, Beltraniella humicola, B. portoricensis, Microxiphium theae, and Piceomphale pinicola. Their morphological characteristics as well as the results of molecular phylogenetic analysis are reported here.

The goal of this study was to isolate and identify the wild yeast strains in intestine of Othius punctulatus collected in Cheongju-si, Chungcheongbuk-do, Korea and characterize their unrecored yeast strains. We isolated 16 wild yeast strains from 5 intestine samples from Othius punctulatus . The 16 wild yeast strains were included three strains in the genus Aureobasidium, four strains in the genus Cystofilobasidium, one strain in the genus Mrakia, three strains in the genus Naganishia , two strains in the genus Saitozyma , two strains in the genus Sampaiozyma and one strain in the genus Scheffersomyces . Among them Cystofilobasidium capitatum YP53, Cystofilobasidium capitatum YP71, Cystofilobasidium macerans YS620, Cystofilobasidium macerans YS622, Mrakia aquatica YP158 and Scheffersomyces stipitis YI5 were recorded for the first time in Korea. The microbiological characteristics and carbon assimilation of these previously unrecorded yeasts were investigated. Almost all of them were oval-shaped. And, glycerol, L-arabinose, xylitol, and inositol are not utilized.

The goal of this study was to isolate wild yeasts from waters and soils in Sangjubo of Nakdong river and Daechung dam in Daejeon city, Korea and investigate characteristics of previously unrecorded novel wild yeasts. In total, 94 strains from 39 different species of wild yeasts were isolated from 47 soils and waters samples obtained from Sangjubo in Nakdong river. Among these, 3 strains, including Rhodosporidium azoricum JSL-GRU-009, represented novel yeast strains which were previous recorded. Two hundred nine strains from 105 different species of wild yeasts were isolated from 130 soils and water samples obtained from Daechungdaegyo and Daechung dam of Shintanjin in Daejeon city, Korea. Ten strains including Bullera pseudoalba JSL-GRU-005 also represented newly recorded strains in Korea. All the 13 previously unrecorded yeasts were oval or global in shape, and nine strains, including R. azoricum JSL-GRU-009, formed ascospores. Ten strains grew well in vitamin-free medium. R. azoricum JSL-GRU-009 grew well in 20% glucose-containing yeast extract-peptone-dextrose (YPD) medium and 3 strains, including Rhodotorula oryzicola JSL-GRU-003, grew well in 5% NaCl-containing YPD medium.

8Diversity and Distribution of Bulb-associated fungi of Fritillaria Cirrhosae Bulbus Source Plants used in Traditional Chinese Medicine

Diversity and community composition of bulb-associated fungi of Fritillaria Cirrhosae Bulbus source plants, which are used in traditional chinese medicine, in the eastern Himalaya- Hengduan Mountains, southwestern China, were estimated based on the internal transcribed spacer rDNA sequence analysis, using host plant species, geographic area, and plant phenology as variables. A total of 1,486 fungal sequences assigned to 251 operational taxonomical units (OTUs) were obtained from the bulbs. Fungal OTUs comprised 96.41% Ascomycotina, 3.52% Basidiomycotina, and 0.07% Zygomycotina. Sordariomycetes, Hypocreales, and Nectriaceae were the most frequent fungal lineages at each taxonomic rank. Fusarium, Ilyonectria, Tetracladium, Leptodontidium, and Tomentella were the top OTU-rich genera. Fusarium sp. 03, Ilyonectria rufa, Fusarium sp. 08, Ilyonectria sp. 03, and Leptodontidium orchidicola 03 represented the most frequent OTUs. Fusarium spp. were the most frequent general taxa. The distribution of fungal community exhibited preferences for host plant species, geographic area, and plant phenology. These findings are the foundation of our research on culturing and active metabolites of bulb-associated fungi of Fritillaria Cirrhosae Bulbus source plants.

Calocybe indica is an edible mushroom commercially cultivated in India and other tropical countries. In this study, the culture characteristics and optimal conditions of milky mushroom strains were determined. The growth temperature and pH range of milky mushrooms was extensively investigated between 15-35 ℃ and pH 3-11. For efficient cultivation, 20 types of nutrient sources were selected that consisted of one of 21 types of carbon sources, 6 organic nitrogen sources, 6 inorganic nitrogen sources, 13 amino acids, 6 organic acids and 12 inorganic salts. The impact of each of the selected nutrition sources and their concentration on growth was investigated. The optimum pH and temperature were determined to be pH 6.0 and 15 ℃, respectively. The optimum concentration of medium elements for the mycelial growth of C. indica was determined to be as follows: carbon source, 2% maltose; organic nitrogen source, 1% yeast extract; inorganic nitrogen source, 0.1% NaNO3; amino acid, 0.7% asparagine; organic acid, 0.07% acetic acid; inorganic salt, 0.07 mM MnSO4.

The goal of this study and potent whitening tyrosinase inhibitor-producing wild yeasts and further optimiz production of tyrosinase. Among non-pathogenic wild yeasts obtained from soils in Daejeon city and spice field of Geumsan in Chungcheongnam-do, Korea, we selected Saccharomyces cerevisiae (S. cerevisiae ) WJSL0191 and Papiliotrema laurentii (P. laurentii ) ON30 show 33.2% and 27.3% respectively. These selected strains formed and not pseudomycelium. S. cerevisiae WJSL0191 was sugar-tolerant as well as halophilic in 20% glucose-containing yeast (YPD) medium and 15% NaCl-containing YPD medium. S. cerevisiae WJSL0191 and P. laurentii ON30 showed 26.2% and 18.6% anti-wrinkle elastase inhibitory activities, respectively. aximal production of tyrosinase inhibitors obtained when S. cerevisiae WJSL0191 was cultured at 30 for 72h in YPD medium and P. laurentii ON30 was incubated at 20℃ for 24hr.
