Objectives : The purpose of this study was to document the use of medicinal plants in traditional practices and to analyze and evaluate medicinal traditional knowledge of indigenous people in Wolchulsan National Park.
Methods : Data were collected through interviews, informal meetings, open and group discussions, and observations guided by semi-structured questionnaires. Data were analyzed via quantitative analysis of use value (UV), informant consensus factor (ICF) and fidelity level (FL), and network analysis.
Results : A total of 580 methods of usage recorded in this study were classified into 55 families, 95 genera, and 104 species. Plants with the highest recorded UVs were Glycine max (L.) Merr., Leonurus japonicus Houtt., and Artemisia princeps Pamp.. The informant consensus factor about using medicinal plants ranged from 0.55 to 0.92, which showed a high level of agreement among the informants on respiratory system disorders and pains. There were 22 species of plants with a fidelity level of 100 %, after eliminating the plants that were mentioned only once from the analysis. Finally, using network analysis, Glycine max (L.) Merr. and Artemisia princeps Pamp. were defined as species with meaningful medicinal use, while lumbago and leg pain were defined as significant ailments in the study area.
Conclusions : This study highlights the diversity and importance of medicinal traditional knowledge for communities of Wolchulsan National Park, Korea. The results of this study will provide basic data for phytochemical and pharmaceutical studies, such as new medicines and therapies.

Objectives : Ephedra regeliana , although similar to the other E. sinica , E. intermedia , and E. equisetina , is not authorized by The Korean Pharmacopoeia and yet has also been imported under the name of 'Ma-Hwang'. The aim of this study was to perform a comparative anatomical evaluation of Ephedrae Sinicae Herba (ESH), Ephedrae Intermedicae Herba (EIH), Ephedrae Equisetinae Herba (EEH) and Ephedrae Regelianae Herba (ERH).
Methods : Permanent paraffin-embedded specimens were produced using the paraffin-tertiary butyl alcohol (TBA) method, to visualize their anatomical features via light microscopy.
Results : ESH and ERH transverse sections were in the shape of a long ellipse, while those of EIH and EEH were in the shape of a circle. These anatomical characteristics substantiated the results of external morphological examination. The sections revealed that each of the four samples showed significantly different cortexes. The cortex of ESH was wider than two thirds of the section, unlike that of the other samples which was relatively narrow. The xylems of ERH, EIH, and EEH demonstrated interconnectivity with other adjacent xylems, contrary to those in ESH. Therefore, the characteristic development of the xylems over time was used to distinguish ERH from the other. Furthermore, parenchymal morphology in the central cylinder was also a good criterion to differentiate between EIH and EEH, since EIH demonstrated parenchymal cells that were shaped like a rounded-triangle, whereas EEH parenchymal cells were chrysanthemum-shaped.
Conclusions : The identification-keys established in this study would facilitate the accurate identification of microscopic features of the four varieties of 'Ma-Hwang'.

3한국 속리산 국립공원 지역집단에서 이용되는 약용동물의 구전 전통지식에 대한 민족동물학적 연구

Objectives : This study aims to record and conserve oral traditional knowledge of medicinal animals from the indigenous people living in the local communities of Songnisan National Park, Korea.
Methods : Data was collected by participatory observations and in-depth interviews with semi-structured questionnaires. Quantitative comparative analyses were accomplished through data received from the following three methods: informant consensus factor (ICF), fidelity level (FL), and network analysis.
Results : The investigation reveals that the indigenous people have used 49 species of medicinal animals distributed within 45 genera, belonging to 39 families with 336 different usages. According to the distribution of recorded families, the most representative families were Scolpendridae and Phasianidae, which were utilized 36 times each (10.71 % each). The category with the highest degree of consensus from informants was disorders related to the nervous system (0.97). 16 species were classified with a fidelity level of 100 %. The network analysis revealed that a lack of vigor was related to 23 species, including Agkistrodon blomhoffii , Gallus domesticus , and Canis familiaris , among the total 49 species investigated.
Conclusions : This documentation can help preserve the traditional knowledge and local health traditions of Korea that are disappearing due to rapid industrialization, urbanization, and death of the elderly with traditional knowledge. Additionally, the animals investigated in this study can be developed into medicinal food and drug for treating specific health conditions through further research.

Objective : Diabetic gastroparesis is a complication that is defined as delayed gastric emptying and upper gastrointestinal symptoms and often occurs in long-standing diabetic patients. Bojungikgi-tang (BJT) is a traditional oriental herbal formula that has long been used for the treatment of digestive disorders. The purpose of this study was to investigate the effects of BJT on streptozotocin (STZ)-induced diabetic gastroparesis rat model.
Methods : Sprague-Dawley (SD) male rats (250-270g) were divided into 13 groups including normal group, STZinduced diabetic control group, BJT diet (7 various concentrations), and insulin-, glibenclamide-, metformintreated group were used for the experiments for the comparison. Diabetic gastroparesis was induced by intraperitoneal injection of STZ. The water intake, food intake, body weights and fasting blood glucose levels were measured. After 4 weeks the animals were sacrificed and gastrin, leptin, insulin, hemoglobin A1C (HbA1c), lactate, lactate dehydrogenase (LDH), bilirubin, creatinine, albumin and lipid levels were evaluated.
Results : Intraperitoneal injection of BJT for 4 weeks resulted in increased levels of gastrin in blood and decreased leptin and lactate concentration in STZ-induced diabetic gastroparesis rat model. BJT did not affect insulin, fasting glucose, HbA1c, and lipid levels in STZ-induced diabetic gastroparesis rat model.
Conclusion : These results indicated that BJT would have protect effect on diabetic gastroparesis through the improvement effect of gastric motility and fatigue syndrome in STZ-induced diabetic rats. This study shows that BJT might be effective for treatment of diabetes and its complications such as gastroparesis.

Objective : Gastritis is a major complication of gastrointestinal disease. Lonicera japonica is used in folk medicine to treat different diseases such as exopathogenic wind-heat, epidemic febrile diseases, sores, carbuncles and some infectious diseases. Therefore, this study examined the effects of Lonicera japonica water extract (LJE) on HCl/ethanolinduced acute gastric ulceration and anti-oxidants properties.
Methods : LC-ESI-IT-TOF MS was employed for rapid identification of major compound from LJE. The antioxidant activities were evaluated through total polyphenol and flavonoid contents and radical scavenging assays and superoxide dismutase (SOD)-like activity. SD rats were randomly divided into five different groups including the normal group, ulcer group, positive group (20 ㎏/㎎ of omeprazole, ip), and experimental groups (100 ㎏/㎎ and 500 ㎏/㎎ of LJE, ip).
Results : 4,5-Dicaffeoyl quinic acid, loganic acid, secologanic acid, sweroside, loganin, vogeloside were identified based on the detection of the molecular ion with those of literature data. The LJE was possessed free radical scavenging activities such as DPPH (IC50=189.7 ㎍/㎖), ABTS (IC50=164.5 ㎍/㎖), and SOD-like activity (IC50=405.02 ㎍/㎖). Macroscopic and histological analyses showed LJE treated group were significantly reduced to an extent that it allowed leukocytes penetration of the gastric walls compared with the ulcer group. In addition, an ulcer inhibition rate and prostaglandin E2 levels were increased in rats treated with LJE.
Conclusion : The present study has demonstrated the antioxidantive and gastroprotective effect of LJE, these findings suggested that LJE has the potential for use in treatment of gastric disorders.

Objective : Arnebiae Radix, the roots of Arnebia euchroma (Royle) Johnst. or A. guttata Bunge, is a well-known traditional herbal medicine in East Asia. In the Korean Pharmacopoeia, three authentic species, including two Arnebia and Lithospermum erythrorhizon Sieb. et Zucc. have recognized as a Lithospermi Radix. But, the morphological and taxonomic information of two authentic Arnebia species lacks date since their distribution and difficulty of accessing the natural habitats.
Methods : A full description of the external morphological characteristics of vegetative and reproductive organs in the original plants was carried out using digital calipers and stereoscope. For providing their taxonomic and nomenclatural synonyms, we reviewed various plant checklists and taxonomic literature.
Results : This study provides taxonomic information of A. euchroma and A. guttata based on a morphological examination of authentic plant individuals and field observations. Detailed descriptions of the two Arnebia species, major quantitative and qualitative characteristics, nomenclatural reviews, distribution maps, and lists of voucher specimens examined are provided. The shape of leaves, root length, stem number, calyx length and pubescence as well as corolla color were useful characteristics for identification. Moreover, we confirmed that A. euchroma and A. guttata are clearly heterostylous species.
Conclusion : Our research provides valuable basic information that could be used as quality control for distinguishing between Arnebiae Radix and its adulterants. These results will help in the understanding of authentic plants and also provide the verified materials and voucher information.

Objective : The purpose of the study was to identify expression profiling of miRNAs associated with cancers after treating allergen-removed Rhus Verniciflua Stokes and allergen-removed Rhus Verniciflua Stokes fumigaed Angelica gigas on leukemia cell lines.
Methods : miRNA expression has been analyzed using miRNA array method through denaturation and hybridization after isolating the total RNA from leukemic cell line treated with 100 ㎍/㎖ of aRVS and aRVS-A each. Microarray expressions were interpreted as 'significant' on miRNAs when decreased less than 0.5 fold or increased more than 1.5 fold compared with the control group.
Results : Among 158 miRNAs in total, 32 miRNAs were significantly presented in miRNAs expression. miRNA has been activated with a variety of genes for predicted targets, and the overexpressed miRNAs were categorized according to proliferation and metastasis of cancer in this study. The findings were reported that seven miRNAs (let-7b, miR-193a-5p, 296-3p, 26a, 22, 124a, 92b) showed significant expressions on proliferation and growth, seven miRNAs (miR-193a-5p, 26a, 200c, 183, 124a, 198, 210) presented meaningful expressions on invasion and metastasis, two miRNAs (let-7b, miR-210) were highly expressed on angiogenesis, five miRNAs (let-7b, miR-26a, 181d, 181c, 296-5p) related with apoptosis, and six miRNAs (let-7b, miR-200c, 183, 370, 124a, 191) were associated with prognosis of cancer and early diagnostic factors for cancer.
Conclusion : The mechanism of miRNA takes a role in diagnosis, treatment, and prognotic factors for cancer as well. This study suggested that further detailed research on overexpression of specific miRNA should be carried out continuously in the future.

Objective : This study investigated the anti-diabetic effects of DM1, a herbal mixture with Atractylodis Rhizoma, Anemarrhenae Rhizoma, and Cinnamomi Cortex in high fat diet (HFD)-induced diabetic mice and the mechanism in C2C12 mouse skeletal muscle cells.
Methods : The C57B/6 mice were fed high fat for 12 weeks, and then administrated DM1 extract (500 ㎎/㎏, p.o.) for 4 weeks. The changes of body weight, calorie and water intakes, fasting blood glucose levels and the serum levels of glucose, insulin, triglyceride, HDL-cholesterol, AST and ALT were measured in mice. The histological changes of liver and pancreas tissues were also observed by H&E stain. C2C12 myoblasts were differentiated into myotubes and then treated with DM1 extract (0.5, 1, and 2 ㎎/㎖) for 24 hr. The expression of myosin heavy chain (MHC), PGC1α, Sirt1 and NRF1, and the AMPK phosphorylation were determined in the myotubes by western blot, respectively.
Results : The DM1 extract administration significantly decreased the calorie and water intakes, glucose, triglyceride, AST and ALT levels and increased insulin and HDL-cholesterol in HFD-induced diabetic mice. DM1 extract inhibited lipid accumulation in liver tissue and improved glucose tolerance. In C2C12 myotubes, DM1 treatment increased the expression of MHC, PGC1α, Sirt-1, NRF-1 and the AMPK phosphorylation.
Conclusion : In our results indicate that DM1 can improve diabetic symptoms by decreasing the obesity, glucose tolerance and fatty liver in HFD-induced diabetic mice, and responsible mechanism is might be related with energy enhancement.

Objective : To establish a reliable tool between for the distinction of original plants of Sanguisorbae Radix, we analyzed the complete chloroplast genome sequence of Sanguisorbae Radix and identified single nucleotide polymorphisms (SNPs).
Materials and methods : The chloroplast genome sequence of Sanguisorba officinalis, Sanguisorba tenuifolia f. alba (Trautv. & Mey.) Kitam and Sanguisorba tenuifolia Fisch. ex Link obtained using next-generation sequencing technology were described and compared with those of other species to develop specific markers. Candidate genetic markers were identified to distinguish species from the chloroplast sequences of each species using Modified Phred Phrap Consed and CLC Genomics Workbench programs.
Results : The structure of the chloroplast genome of each sample that had been assembled and verified was circular, and the length was about 155 kbp. Through comparative analysis of the chloroplast sequences, we found 220 nucleotides, 158 SNPs, and 62 Indel (insertion and/or deletion), to distinguish Sanguisorba officinalis, Sanguisorba tenuifolia f. alba (Trautv. & Mey.) Kitam and Sanguisorba tenuifolia Fisch. ex Link. Finally, 15 specific SNP genetic markers were selected for the verification at positions. Avaliable primers for the dried herb, which is used as medicine, were used to develop the PCR amplification product of Sanguisorbae Radix to assess the applicability of PCR analysis.
Conclusion : In this study, we found that Fendel-qPCR analysis based on the chloroplast DNA sequences can be an efficient tool for discrimination of Sanguisorba officinalis , Sanguisorba tenuifolia f. alba (Trautv. & Mey.) Kitam and Sanguisorba tenuifolia Fisch. ex Link.

10모과추출물의 C2C12 근육세포에서 근분화 및 에너지대사조절인자 발현 증진 효과 연구

Objective : The present study was conducted to investigate the effects of Chaenomelis Fructus (CF) on the regulation of biogenesis in C2C12 mouse skeletal muscle cells.
Methods : C2C12 myoblasts were differentiated into myotubes in 2% horse serum-containing medium for 5 days, and then treated with CF extract at different concentrations for 48 hr. The expression of muscle differentiation markers, myogenin and myosin heavy chain (MHC) and mitochondrial biogenesis-regulating factors, peroxisome proliferatoractivated receptor gamma coactivator 1alpha (PGC1α), sirtuin1 (Sirt1), nuclear respiratory factor1 (NRF1) and transcription factor A, mitochondrial (TFAM), and the phosphorylation of AMP-activated protein kinase (AMPK) and acetyl-CoA carboxylase (ACC) were determined in C2C12 myotubes by reverse transcriptase (RT)-polymerase chain reaction (RT-PCR) and western blot, respectively. The cellular glucose levels and total ATP contents were measured by cellular glucose uptake and ATP assays, respectively.
Results : Treatment with CF extract (0.01, 0.02, and 0.05 ㎎/㎖) significantly increased the expression of MHC protein in C2C12 myotubes compared with non-treated cells. CF extract significantly increased the expression of PGC1α and TFAM in the myotubes. Also, CF extract significantly increased glucose uptake levels and ATP contents in the myotubes.
Conclusion : CF extract can stimulate C2C12 myoblasts differentiation into myotubes and increase energy production through upregulation of the expression of mitochondrial biogenetic factors in C2C12 mouse skeletal muscle cell. This suggests that CF can help to improve skeletal muscle function with stimulation of the energy metabolism.
