We examined the relationship between the basic characteristics and types of leisure activities according to blood types. It was based on questionnaires answered by 441 collegians in public health departments and 282 in non-public health departments in Daegu Health College in March, 2005. The frequency of leisure activity of once or twice a week or less during weekdays was the most common.: The most common frequencies for all blood types were between 1 and 2 hours on weekdays and between 3 and 5 hours on the weekends or holidays. As far as the type of leisure activities during the weekdays was concerned, in the case of public health departments, blood type A was interested in computer games and then watching TV, blood type B was attracted to watching TV and listening to music, blood type AB was into watching TV, computer games, and listening to music, and blood type O was keen on watching TV and computer games in that order. The type of leisure activity according to blood types in public health departments was shown to be significantly different (p<0.05). The type of future leisure activity was shown to be significantly different between public health departments and non-public health departments (p<0.05). The type of present leisure activity preferred by collegians, according to the study, was entertainment such as computer games for most blood types. The type of future leisure activities was related to the field of tourism and sports. The collegians need to have more advanced and positive leisure activities.

The purposes of this study were to evaluate the changes in hematologic indices after plateletpheresis and to identify the preapheresis platelet count and clinical factors (age, gender, height, and weight) that showed some influence on the percentage of platelet decrement, yield and efficiency. Plateletpheresis was performed on 101 healthy donors in Bundang CHA general hospital. The data was analyzed using the SAS program with t-test, ANOVA test and Multiple regression. The mean percentage decrease after plateletpheresis was 2.0% in hemoglobin, 1.8% in hematocrit, and 29.7% in the platelet count, while a WBC count showed an increase of 2.6%. The mean percentage decrease of hemoglobin and hematocrit were 1.7% and 1.4%, in males and 3.6% and 3.7% in females, respectively. Particularly the percentage decrease of platelet count was significantly higher in females (40.0%) than in males (27.2%). The platelet decrementage and yield were significantly higher in females, but the efficiency did not differ significantly between males and females. The yield showed the lowest levels in subjects who were 40 years old or over but the platelet decrement and efficiency did not change according to age. The platelet decrement increased as height and weight increased. Also, the platelet decrement and yield increased as the initial platelet counts increased, but the efficiency did not. From multiple regression analysis, the platelet decrement was associated with gender, weight, and initial platelet count. The yield was related to the initial platelet count, but the efficiency was not related to gender, age, weight, height or initial platelet counts. This study has a limitation of the generality of the study results since this study was conducted only in a single university hospital. Further study would be necessary to find out a subpopulation that is sensitive to the hematologic change after plateletpheresis, and to determine the standard criteria for blood donation based on the subpopulation.

Peripheral bood stem cell collection (PBSCC), including peripheral blood stem cell transplantation (PBSCT), has been utilized worldwide as a very beneficial treatment method instead of allogenic Bone Marrow Transplantation (BMT) because it has many advantages such as rapid bone marrow engraftment and hematopoietic recovery, easy and safe accessibility and lower risk of rejection compared with allogenic BMT. In order to identify most the observable parameter in PBSCC, we analyzed various hematological parameters before and after PBSCC, and evaluated the correlation between the time of bone marrow engraftment and the number of CD34+ cells. Thirteen patients, who underwent 54 PBSCCs from January, 2003 to August, 2004 at Chungnam National University Hospital due to various systemic neoplasms, were analyzed in aspects of various hematological parameters including CD34+ cells using by Flow Cytometry (FCM). PBSCC harvests are described below: Mononuclear cells (MNC) 2.3 ± 1.4×108/kg and CD34+ cells 0.63 ± 0.35×106/kg on average, respectively. There was a statistical significance in Hb and Hct before and after PBSCC, but not in WBC and platelet counts. The period to reach the hematological bone marrow engraftment was 13.4(10~21) days and 19.5(11~38) days according to the criteria of absolute neutrophile counts (ANC) ≥ 500/uL and platelet counts ≥50,000/μL in peripheral blood, respectively. There was a significant correlation between the numbers of CD34+ cell and ANC (p<0.05), and a borderline significance between MNC and ANC (p=0.051). We found that a group of patients, who were infused with CD34+ cells more than 3.5×106/kg, reached more rapidly the period of bone marrow engraftment in platelet counts (p=0.040). This present study suggested that Hb and Hct were the most useful parameters and should be closely monitored before and after PBSCC, that a PBSCT with the dosage of more than 3.5×106/kg of CD34+ cells was needed to perform successful bone marrow engraftment, and additionally that platelet counts could be more useful in indicating bone marrow engraftment than ANC.

Parallel testing means ordering a number of tests at the same time so abnormalities in any of the tests can be found quickly and used in making the diagnosis. This is a good medical strategy to eliminate diseases and it is relatively inexpensive if all the tests are potential sources of information and performed on the same analyzer. In regression, the equation for the straight line is recast as y = bx + a. This change in terminology leads to confusion. Here a is the y-intercept or constant and b is the coefficient or slope of the line. A few more words of caution about regression - as in all of statistics there are certain assumptions: the x value is a true measure, both X and Y distributions are normal, and homoscedasticity, i.e., the variance of y is the same for each value of x. In this study the linearity classification made by different scientists were always in agreement. Typical examples of curves that were considered linear are presented in Fig. 1-5. Because these automated procedures values were usually within five percent of each other the curvature could be easily detected. The plot of the WBC, RBC, hemoglobin, hematocrit and platelet concentrations from approximately 74.4 to 0×103/μL and 80.4-0×103/μL, 5.6-0×106/μL and 6.1-0×106/μL, 18.3-0 g/dL and 19.0-0 g/dL, 54.1-0% and 56.8-0% and 642.0 to 0.03×103μL and 754.0-0×103/μL on the ADVIA 2120 C Versus and A and B typical of an acceptable linear study as shown in Fig. 1-5. The grand mean of R2, intercept and slope is 0.99898, 0.99459 and 1.54626.

Ochrobactrum anthropi, previously known as Achromobacter species biotypes 1 and 2 (CDC groups Vd-1, Vd-2), belong to the groups of non-Enterobacteriaceae- nonfermentative Gram negative bacilli. Achromobacter is not presently a recognized genus. Achromobacter xylosoxidans has been transferred to genus Alcaligenes as A. xylosoxidans subsp. xylosoxidans, and "Achromobacter" sp. group Vd has been named Ochrobactrum anthropi. O. anthropi was isolated from a blood culture. Organisms were identified as O. anthropi by use of the biochemical test and the VITEK 2(bioMerieux, USA). The Organism was susceptible only to colistin, imipenem, meropenem, and tetracycline, but were resistant to amikacin, aztreonam, cefepime, ceftazidime, cefpirome, ciprofloxacin, gentamicin, isepamcin, netilmicin, pefloxacin, piperacillin, piperacillin/tazobactam, ticarcillin, ticarcillin/clavulanic acid, tobramycin, and trimethoprim/sulfamethoxazole. We report the clinical and microbiologic characteristics of O. anthropi infection in the patient. This is the first case of O. anthropi infection after using a plant as medicine at Chosun University Hospital.

Recently, the rapid increase in extended-spectrum β-lactamase (ESBL) producing clinical isolates has become a serious problem. In this study, the epidemiologic features and molecular characteristics of ESBL among clinical isolates of Escherichia coli and Klebsiella pneumoniae, antibiotic susceptibility testing, genotype of the ESBL and patterns of chromosomal DNA from PFGE (pulsed field gel electrophoresis) were observed. A total of 53 ESBL-producing clinical isolates (30 of E. coli and 23 of Klebsiella pneumoniae) were collected from two university hospitals in the period of June to July in 2002 and 2003 respectively. The antibiotic resistance frequency of those 53 strains was tested by the disk agar diffusion method with the result that all the strains were resistant to cephalothin. To other antibiotics, the resistance rates of E. coli (30 isolates) were in order of ceftazidime (90.0%), cefotaxime and aztreonam (respectively 83.3%). Also, the resistance rates of K. pneumoniae (23 isolates) were in order of aztreonam (78.3%), ceftazidime (73.9%) and cefotaxime (65.3%). Also the sensitivity of ceftazidime-clavulanic acid were 100% in E. coli and 95.7% in K. pneumoniae. And the sensitivity of cefotaxime-clavulanic acid was 96.7% in E. coli and 91.3% in K. pneumoniae. The types of the ESBL genes were determined by using polymerase chain reaction (PCR). Among the 30 isolates of ESBL-producing E. coli, 6 (20.0%) have SHV only, 5 (16.7%) have TEM only and, 18 (60.0%) have both of TEM and SHV. Among the 23 isolates of ESBL-producing K. pneumoniae, 7 (30.4%) have SHV only, 2 (8.7%) have TEM only, and 14 (60.9%) have both of TEM and SHV. These results show that 52 strains, with only one exception, were confirmed as either TEM or SHV. The patterns of XbaⅠ-digested chromosomal DNA of ESBL-producing E. coli and K. pneumoniae isolates were analyzed by PFGE. PFGE patterns of E. coli and K. pneumoniae were multiclonal, but many strains were grouped into a few types. Therefore, it seems that there were clonal outbreaks or possible horizontal spread. In conclusion, the TEM and SHV β-lactamase are most widely spread in E. coli and K. pneumoniae in Korea. As these types are usually carried by plasmids, the spread of these β-lactamase genes could compromise the future usefulness of third generation cephalosporins for the treatment of infections caused by E. coli and K. pneumoniae.

Beta-hemolytic streptococci (BHS) are frequently isolated pathogens in clinical microbiology laboratories. Among them, Group C, G and F streptococci infrequently cause respiratory infections, but they often invade other sites. Patients with underlying diseases are prone to be infected by the organisms and some of them can be fatal. Therefore, combination tests of serological and biochemical tests are needed to correct the identification of non-A, B streptococci because to various species belong to this species. The aim of this study was to investigate the isolation rates of strains and distribution of serogroup C, G and F streptococci at Wonju Hospital during the period of 2003-2004. 133 clinical isolates of group C, G and F streptococci were examined. Of them, 13 (9.8%) were group C, 41 (31.8%) were group F, 79 (59.4%) were group G. The prevalent isolation of the strains by serogroup were: group C, Streptococcus constellatus (38.5%); group F, Streptococcus anginosus (56.1%), Streptococcus constellatus (26.9%); group G, Streptococcus anginosus (54.4%), Streptococcus dysgalatiae subsp. equisimilis (26.5%). Overall, the most frequently isolated BHS was Streptococcus anginosus (51.2%).

A simple and easy modification of AST by disk diffusion was tested for the detection of induced clindamycin resistant Staphylococci and their antimicrobial susceptibility at the same time. The incidence of inducible clindamycin resistant staphylococci in blood culture and their MIC characterization at Seoul National University Hospital was analyzed by an AST contained disk approximation test (D-zone test) and Etest, respectively. Of the total 309 staphylococcal isolates, 139 (45%) isolates presented constitutive resistance to ERY and CLI (ERY-R, CLI-R phenotype), and 59 were ERY-I/R and CLI-S phenotypes. Of the 59 isolates, 19 (32%) isolates were inducible resistant to CLI. The incidence was higher in S. aureus (66.7%) than coagulase-negative staphylococci (CNS, 26.0%). Especially, methicillin-resistant staphylococci (MRSA, 100%; MRCNS, 45.5%) presented higher inducibility than methicillin susceptible (MSSA, 50%; MSCNS, 20%). For most of the inducible clindamycin resistant staphylococci (15 of 19 isolates), their ERY MIC were high (>128 μg/mL) and were methicillin resistant. The remaining 4 isolates were methicillin susceptible and their ERY MIC were of intermediate concentrations (1-4 μg/mL). We concluded that suscetibility testing of staphylococci, especially methicillin resistant, should include the D-zone test.

Anaerobic black-pigmented bacteria have been implicated in the endodontic infections. This group of microorganisms includes Porphyromonas endodontalis, P. gingivalis, Prevotella intermedia, and Prevotella nigrescens. The organisms display a wide variety of virulence factors that may be pertinent to acute endodontic infections. The aim of this study was to identify P. endodontalis, P. gingivalis, P. intermedia, and P. nigrescens by using the special potency disk test, filter paper spot test, 16S rRNA gene-directed PCR, and API 32A system. Microbial samples were collected from root canals of 33 intact teeth with necrotic pulp and apical periodontitis. Conventional laboratory methods were used to identify the strains of anaerobic black pigmented bacteria. Eighteen out of 33 samples were positive for the growth of black-pigmented bacrteria. Five colonies were cultured from each pure cultured colony from Brucella agar plates. Seventy seven colonies were positive for the growth of black-pigmented bacteria. Thirty three out of 77(42.8%) were identifed as P. nigrescens, 10 out of 77(13%)were P. gingivalis, 6 out of 77(7.8%) were P. endodontalis, 10 out of 77(13%) were P. intermedia. On the contrary the reference strains of P. nigrescens, experimental strains of P. nigrescens were susceptible to kanamycin in the special potency disk test. We concluded that after rapid presumptive identification methods, such as the special potency disk test and filter paper spot test were done, 16S rRNA gene PCR and API 32A test would be accurate detection methods for black-pigemented bacteria.

To find out the effectiveness of clinical examination for the diagnosis of respiratory tuberculosis, a 78 respiratory tuberculosis patient,s group was matched by sex and age with 78 control healthy subjects. In the result of blood chemistry, mean values of 123.5 ± 62.04 mg/dL in glucose, 429.01 ± 150.77 IU/L in LDH, and 44.51 ± 43.76 IU/L in γ-GTP, were higher than that of the controls (healthy subjects), and 3.51 ± 0.68 mg/dL in albumin was lower than that of the controls. In the result of the haematology examination, mean values of 12.52 ± 3.27 g/dL in hemoglobin, 36.72 ± 7.28% in hematocrit, and 24.61 ± 12.36% in lymphocyte, were lower than that of the controls, 9.23 ± 5.25% in monocyte 78.30 ± 37.35 mm/hr in ESR, and 48.45 ± 35.15 U/L in ADA were higher than that of the controls. For the comparisons of the tuberculosis patients values from normal reference values, 22.2% in glucose, 22.4% in LDH, 25.0% in γ-GTP, 35.4% in albumin, 88% in ESR, and 88.6% in ADA, showed abnormal values. We concluded that the values of glucose, γ -GTP, albumin, WBC, RBC, hemoglobin, hematocrit, lymphocyte, monocyte, ADA, and the ESR were useful in tuberculosis diagnosis.

The pH-ISE(ion selective electrode) based on tribenzylamine as a hydrogen ion carrier was prepared and its electrochemical characterization was studied. It responded linearly to hydrogen ions in the range of pH 3.1 - pH 11.0 and the Nernstian slope showed 55.0 mV/pH (at 20 ± 0.2℃), it also showed a fast response time of 8 sec. When it was directly applied to human blood(pH 6.0-8.5), we could get the same satisfying results. A good reproducibility and stability were shown with the precision of 2 mV (±0.1). The pH-ISE based on tribenzylamine exhibited biocompatibility in clinical applications.
